VITROCELL® is specialised in the development of advanced in vitro exposure systems. VITROCELL© realizes turnkey installations for in vitro inhalation toxicology in which gases, environmental atmospheres, nanoparticles and complex mixtures are analyzed on lung cells at the air/liquid interface.

Setup

The VITROCELL®  Exposure System consists of an aerosol generator selected for the optimal aerosolization of substances. It includes the possibility to vary doses by dilution. Then an aerosol sample stream is guided to the exposure module which consists of an exposure top with specially designed inlets and a heated base module to receive the cell culture media and cells which are cultivated on cell culture inserts. According to the capacity needs there is a choice of using inserts in the 6, 12 or 24 well format. Exposure can take place from 3 up to 24 inserts at stable and individually controlled conditions.

暴露染毒系统由发生器(根据研究方向不同选择合适的发生器)、稀释系统、染毒模组组成。其中染毒模组内可放置61224孔板培养皿以满足实验需要。每套系统内的染毒槽从3个到24个不等。

Air-Liquid Interface (ALI) exposure

气液界面暴露技术

In the early phases of testing airborne materials in vitro scientists had no other choice than to use a submerged exposure process. Here, the test substances were added to the cell culture media or for gaseous compounds the substances were bubbled through the media using impinger-like devices. Particles were extracted from filters or applied directly by suspension to the media.

When the first cell culture inserts became available, cells could be cultivated on porous membranes and then be exposed at the air-liquid interface – here the cells are not covered with culture media during exposure. This procedure has the advantage that all constituents of the test atmosphere, thus gas, semi- volatile and particle phase can reach the cells directly and in their original composition. So the air-liquid interface enables the testing of substances without changing their original properties. Nutrition of the cell cultures is maintained at the same time through the pores of the membrane of the cell culture insert.

 

在传统的暴露染毒实验中,由于技术受限,只能采用混合模式(培养液与细胞混合)暴露染毒。

而气液界面暴露技术突破了局限,真正实现了对人体环境的真实模拟。在该技术下,细胞被培养在TRANSWELL内贴壁生长,而培养液不在与细胞接触,因此细胞得以在气体、半挥发物质、颗粒物中进行完全暴露,而且受试物质不会因为其他外力改变其性质。实验结果更精准。

Dosimetry via Quartz Crystal Microbalances

 

The novel microbalance system can be integrated in the module for use before the experiment for validation purposes of the aerosol delivery or during the experiment as process control.

The direct reading gives an exact online picture of the exposure situation and delivery of the test substance.
